Dr. John Mancuso is the Director of Chemistry at Sygnature Discovery, leading global peptide and macrocycle synthesis programs and advancing ADC linker–payload innovation for pharmaceutical and biotech partners. His expertise spans synthetic organic chemistry, peptide design, and complex molecule development.

John has co-invented 14 patents and authored more than 15 scientific publications. At Sygnature, he manages multidisciplinary drug discovery projects, integrating chemistry and biology to deliver novel intellectual property and next-generation therapeutics that drive progress in peptide and ADC research.
